Shivanand R. Patil Cytogenetics and Molecular Laboratory

The Shivanand R. Patil Cytogenetics and Molecular Laboratory is located within the Division of Medical Genetics. The lab is a nationally accredited, CAP-inspected, CLIA certified facility that tests and analyzes subtle changes in genes and chromosomes to assist in the diagnosis of complex developmental problems in children and adults using state-of-the-art methods. The laboratory performs conventional chromosome analysis, fluorescence in situ hybridization (FISH), and chromosomal microarrays for a myriad of indications including cancer diagnosis/prognosis as well as both post and pre-natal genetics.

Position Summary

Within the Stead Family Department of Pediatrics- Shivanand R. Patil Cytogenetics and Molecular Laboratory, this position performs components of cytogenetic and molecular cytogenetic studies from biological specimen for diagnosis, prevention or treatment of disorders. This position will work to maintain and improve the testing environment, 

capabilities and quality of data in the Pediatric Cytogenetics Laboratory.

Position Responsibilities

·Perform quantitative and qualitative analysis for all levels of testing complexity on laboratory clinical biological specimens such as blood, bone marrow, tissue and body fluids.

·Perform cell culture on varieties of tissues received for chromosome analysis according to established protocols.

·Process cultured cells and prepares slides according to laboratory protocol of chromosome spreads.

·Stain slides utilizing a variety of techniques including G-banding and fluorescent in situ hybridization.

·Prepare microscope analysis of metaphases and prometaphase chromosomes for diagnostic purposes.

·Prepare digitized karyotypes using correct International System for Human Cytogenetic Nomenclature (ISCN).

·Perform DNA extraction.

·Perform DNA quality control and gel electrophoresis.

·Prepare and process DNA for molecular applications such as polymerase chain reaction (PCR).

·Process DNA for chromosomal microarray (CMA) applications including DNA fragmentation and labeling, hybridization to microarray, microarray washing, and microarray scanning.

·Process DNA for next-generation sequencing (NGS) applications including library preparation, DNA sequencing, initial data analysis.

·Begin and ensure proper execution of analytic pipelines for both CMA and NGS applications.

·Initiate CMA and NGS data analysis pipelines by quality controlling raw data, placing raw data in appropriate file locations to begin analysis, starting analysis, checking quality control metrics as analysis proceeds to ensure proper execution.

·Maintain accurate and legible records of all lab results and procedures, and accurately interprets and records test results.

·Document all specimen information during accession process.

·Investigate quality assurance issues involving patient samples.

·Recognize basic problems including culture failure and contamination as well as DNA extraction failure, fragmented DNA, quality control metrics outside acceptable ranges, poor CMA and/or NGS data quality.

·Recommend corrective action for quality assurance.

·Seek technical direction from laboratory supervisor with specific instrumentation problems.

·Assist in reagent comparisons, evaluate results and recommend changes to reduce costs and improve efficiency.

·Perform daily operation and routine maintenance, calibration, and assist in troubleshooting of laboratory instruments such as incubators, microscopes, hoods, microarray wash stations and scanner, centrifuges, DNA sequencer, PCR thermocyclers, gel electrophoresis equipment.

·Participate in evaluation of equipment and recommend products for acquisition.

·Maintain inventory and orders supplies as needed.

·Comply with applicable regulatory and accreditation requirements of the laboratory section.

·Perform, record and review daily quality control associated with assigned testing.

·Document and report any variances.

·Adhere to quality standard procedures for high complexity testing facilities.

·Participate in the Quality Assurance program.

·Identify opportunities for improvement and make recommendations for resolution of identified problems/errors.

·Maintain quality control and quality assurance procedures in accordance with established policies and regulatory requirements; assemble quality control data for further analysis; recognize problems; and document issues; and initiate corrective actions after reporting problems.

·May assist with writing and revising laboratory procedures according to NCCLS guidelines.

·Assist in a positive and professional manner with bench training for other laboratory colleagues within the section.

·Participate in lab section meetings with others to discuss quality testing, workflow, efficiency strategies, problematic areas, and laboratory improvement.

·Maintain effective working relationships with faculty, staff, students and the public.

·Ability to work with a variety of individuals and groups in a constructive and civil manner while appreciating the unique contribution of individuals from varied cultures, nationalities, genders, ages, etc.

·Ability to utilize existing resources and learning to achieve or exceed desired outcomes of current and future organizational goals/needs. Able to demonstrate ethical behavior in diverse situations while producing results.

Education Requirements

  • Medical Laboratory Technician (MLT) or a categorical/specialist certification in the appropriate laboratory discipline or eligible from American Society for Clinical Pathologists (ASCP) or National Credentialing Agency (NCA), or:
  • Successfully completed an official US military medical laboratory procedures training course of at least 50 weeks duration and have held the military enlisted occupational specialty of Medical Laboratory Specialist (Laboratory Technician) is required.

Experience Requirements

  • Greater than two years of demonstrated experience working in either a clinical laboratory and/or a molecular research laboratory
  • Greater than two years of demonstrated experience with project coordination including time management, decision making, and self-direction in biology, chemistry or allied health or a similar discipline.
  • Excellent written and verbal communication skills.

·Selected candidates will be expected to successfully pass the ASCP Cytogenetics or Molecular Biology Technologist Certification exam within two years from the date of hire.

Desired Qualifications


  • Demonstrated experience with DNA extraction using established protocol.
  • Knowledge of FISH techniques and/or use of molecular probe systems.
  • Knowledge of chromosomal microarray (CMA) and/or next-generation sequencing (NGS) technologies.
  • A Bachelor’s degree in a chemical, biological, or clinical laboratory sciences or related field is highly desirable.
  • Certification from American Society for Clinical Pathologists (ASCP) or National Credentialing Agency (NCA) as a Medical Laboratory Technician (MLT) is highly desirable.
